Shape identification

The capsule is spherical or oblong, the base is blunt, and the longitudinal line of the back suture is straight. The seed is slightly wider than long and heart-shaped. It is better to use a giant, large, yellowish-green, and not broken mantle.

Microscopic identification

1. Fruit flap cross section

In the septum, the cells are elongated, with large wall holes, and the other layer is rectangular or round in shape.

2. Seed cross section

The outer layer of the seed coat is a column of brown cells, with one inlaid wood cell inwards and a square crystal.

Physical and chemical identification

1. The product alcohol leaching solution drops on the filter paper. Observed under the UV lamp, showed yellow-green fluorescence.

2. TLC to take the product powder 3g, add ethanol 50ml, set on a water bath heating 1h, filtered. The filtrate was evaporated to dryness, and the residue was dissolved with 5 ml of ethanol to be used as a test solution, and ethanol solution of aristolochic alcohol was also taken as a control solution. Three μl of each of the above two solutions was spotted on the same silica gel G plate. Chloroform-acetone and acetic acid (8:5:0.1) were used as the developing agent, taken out, dried, and placed on the fluorescent lamp for examination. The same color was spotted on the corresponding positions of the sample and the reference substance.

Notice:

2. Dissolve Proteinase K (20mg/ml): Add Proteinase Dissolve Buffer to dissolve Proteinase K to a final concentration of 20mg/ml. Proteinase K dry powder can be stored at 2-8°C for one year, but dissolved Proteinase K must be stored in aliquots at -20°C. Repeated freezing and thawing of Proteinase K can affect its activity.

3. Buffer VHB must be diluted with 14 ml absolute ethanol before use and stored at room temperature.

4. Buffer RW2 must be diluted with 80 ml of absolute ethanol before use and stored at room temperature.

3. Shelf life

Except for Proteinase K and Carrier RNA, other components of this product can be stored at room temperature (15-25°C) for 12 months, and should be stored at 2-8°C for long-term storage. Proteinase K and Carrier RNA dry powder are transported at room temperature. Please store at -20°C after receiving the test product, and store at -20°C after dissolving.
