AquaMax microplate washer and LifeSep magnetic frame can be applied to MAGPIX multi-magnetic bead detection platform

Introduction:
It is well known that the MAGPIX multiple magnetic bead detection system has high-throughput multi-channel detection characteristics, which can yield 4,800 analysis results in one hour. In order to ensure the best test results, the key step of the test optimization is how to minimize the loss of magnetic beads in the microplate during the cleaning process, thereby improving the cleaning efficiency of the MAGPIX magnetic beads. Molecular Devices' AquaMax2000/4000 microplate plate washer can effectively reduce the loss rate of magnetic beads in the microplate. According to the optimized setting of the instrument, the magnetic beads in the microplate are lost. The rate will be less than 5% (Figure 1).

  • MAGPIX Calibration Kit ( Life Technology, Cat.#MPX-CAL-K25 )
  • Phosphate buffer ( PBS )
  • LifeSep 96F Magnetic Stand ( Dexter Magnetic Technology, Cat.#2501008-1 )
  • 96- well fully transparent microplate ( Greiner )
  • ® Velos TM   High Connotation System ( Molecular Devices )

method:

Step 1: Dilute the magnetic beads in PBS in a ratio of 1:50 , and add 100 ul of magnetic bead mixture to each well of a 96- well plate.

Step 2: Place the microplate on ImageXpress Velos before washing to obtain the number of beads.

Step 3: Set up the AquaMax microplate washer: create a new microplate parameter in the software, select Greiner 96 well plate, height input 24.2mm ( normal microplate height + 10mm LifeSep magnetic frame height ) , use the instrument Self-contained microplate depth detection function, which can accurately detect the depth of the microplate and save the detection data in the program.

Step 4: Use PBS to clean the microplate on the AquaMax4000 washer. After the panel is set up according to the following parameters, the best results will be obtained (effectively reducing the loss of magnetic beads during the cleaning process).

Aspiration

  • Location: Edge
  • Shampoo aspiration rate: medium speed
  • Washing head falling speed: 3
  • Height: 5mm

Packing

  • Volume: 150ul
  • Shampoo dispensing rate: 5
  • Repeat 2 times from the first step
  • Aspiration (set as above)

The green fluorescent signal was scanned using a 532 nm laser source with a ImageXpress Velos high- intensity system . After software analysis, the number of beads in each well before and after microplate cleaning was obtained.

in conclusion:

Effectively reducing the loss rate of the magnetic beads during the cleaning process is a key factor in ensuring the success of the MAGPIX test. When the AquaMax microplate washer cleans the MAGPIX magnetic beads in the 96- well plate , use the LifeSep magnetic frame to perform the instrument as described above. After setting, more than 95% of the beads remain in the microplate. The AquaMax microplate washer has a high degree of auto-adjustment and shampoo interchangeability , ensuring flexibility in adapting to various test requirements.
