NGS gene library construction competition: man-machine war

With the development of sequencing technology, the scientific community has begun to apply more and more sequencing technology to solve biological problems. In the past decade, the next-generation sequencing technology, Second Generation Sequencing (NGS), has grown rapidly, and more and more sequencing laboratories use NGS technology as the primary means of sequencing. In the NGS process, the construction of the gene library is the first step in the whole sequencing process, and the most critical step. The quality of the gene library directly affects the subsequent sequencing work. Although the second-generation sequencing technology is developing rapidly, the construction of the gene library is still a very time-consuming and repetitive work, requiring skilled scientific and technical personnel to maintain high attention throughout the operation, spending 5 hours or more. More time to complete the database construction work.

In order to ease the burden on the sequencing staff and liberate the hands, Roche Diagnostics of France and PrimaDiag announced the development of the ACSIA NGS LibPrep automated pipetting station to provide automation solutions for Roche's library products. The tedious and lengthy construction of the library is automated.

ACSIA NGS LibPrep automatic database system is accurate and repeatable, avoiding any human error during the database construction process, thus improving the quality of the library. Roche Diagnostics offers KAPA library kits for building libraries that are fully automated on this platform without any human involvement. Therefore, using ACSIA NGS LibPrep to build a database is a very economical and cost-effective method. Kapa Biosystems and Roche NimbleGen's database building steps are optimized on the ACSIA NGS LibPrep automation platform, and the running program is preset. Therefore, to build a gene library using the ACSIA NGS LibPrep automation platform, you only need to open the preset method and input. The number of samples, click OK. Manually completing the construction of the library for up to 5 hours, using the ACSIA platform only requires manual operation for 15 minutes, and the rest are replaced by the ACSIA platform.

The ACSIA NGS LibPrep platform uses the innovative patented APIM (Inductive Magnetization Attraction Technology) from Pym . Digg , which generates magnetic depolarization when energized or de-energized according to demand. This minimizes the actual reagent consumption used to build the library, and the optimized operating steps allow the entire library to be completed with a minimum number of tips, saving significant reagent and consumable costs. Building 48 DNA libraries on the ACSIA NGS LibPrep platform requires only 3 cartridges, while the number of tips used for manual operations can reach as many as 14 cartridges; the average number of tips used to process each sample is 28 for manual completion. Comparing the quality of ACSIA and manually established libraries, using ACSIA to process samples, the sample concentration was more uniform and the coefficient of variation was small. In addition to Roche and Kapa Biosystems reagents, Pema Dige has worked with several NGS reagent manufacturers and optimized them, including Illumina, NEB, and Agilent. Figure 1 shows the use of KAPA library kit, using ACSIA automation platform and manual operation to process 12 samples in parallel. The sample concentration of the automated platform is from 22.93 to 27.83 ng/μL, and the coefficient of variation is 1.25. The sample concentration range from the library was 22.52 t to 47.73 ng/μL, and the coefficient of variation was 8.29.
